It has been my experience with several doors that a hard to turn key is related to the latch mechanism.
You end up trying to turn the key against a dirty/rusty or gummed up latch mechnism.
If you remove the inner door panel...use the below thread for help...

...you will be able to access the latch mechanism and the lock cylinder.
When you get into the door, disconnect the rod from the back of the door lock cylinder and then try the key, if it now turns easily I would put my money on the dirty latch mechanism.
If you remove the latch mechanism, (rods on the inside and 3 torx screws on the outside), clean all the old grease and dirt buildup off and regrease it, you will be able to tell the difference in how easy it works compaired to when it was dirty.
I also took the rod off that runs from the inside door handle back to the latch mechanism, the area where it runs through the plastic guide I cleaned with a scotch brite pad to smooth off the roughness, a little grease in the guide and reinstall the rod.
The trouble with the doors is they are not weather tight and water and road dust get in and really gum things up.
If that's not the whole problem changing the lock cylinder out is the next step.
It's not the easiest because of the tight area you have to reach into, like indiana_resto_guy said, small hands help.
There is a c-clip that slides away from the cylinder, it's a pain to remove and replace because it's a tight fit.
I recall using a small channel lock and needle nose vise grips, some screwdrivers and alot of really fowl language directed towards the GM engineers.
Take it slow and you'll get it. LMK if you need any pics of specific areas and I'll see what I can do.
